葛根素对糖尿病大鼠肾组织中CXCL10/CXCR3表达影响的实验研究

摘要: 目的:观察葛根素对糖尿病(Diabetes Mellitus, DM)大鼠肾组织病理形态、肾组织中趋化因子CXCL10以及受体CXCR3表达的影响,探讨葛根素延缓DM大鼠肾组织损伤的机理。方法:采用链脲佐菌素制备DM大鼠模型,将造模成功后的大鼠随机分为DM组(n = 11)、DM组+葛根素干预组(下文简称葛根素组) (n = 11),并设置正常对照组(n = 7)。每2周测定一次各组大鼠空腹血糖、体重。4周后处死大鼠并取大鼠肾脏,对肾脏进行相关处理后,在光镜和电镜下观察大鼠肾组织病理改变,通过RT-PCR检测肾组织CXCL10/CXCR3mRNA的表达水平。结果:在电镜下显示:相较于DM组,葛根素组大鼠肾组织结构破坏程度明显减轻。RT-PCR结果显示:相较于DM组,葛根素组大鼠肾组织中趋化因子受体CXCR3的表达水平明显降低(p < 0.05),但CXCL10的表达水平无明显降低(p > 0.05)。结论:葛根素可能通过降低肾组织中CXCR3的表达水平减轻肾组织的炎症反应,延缓DM大鼠肾组织的损伤。
Abstract: Object: To observe the kidney pathological changes and the expressions of CXCR3/CXCL10 in kidney of diabetic rats under the effect of Puerarin. Methods: Diabetes Mellitus (DM) model rats were established by injecting streptozocin (STZ). Then the successful modeled rats were re-divided into two groups: the DM group (n = 11) and the Puerarin group (n = 11). The normal group (n = 7) was added too. The rats’ fasting plasma glucose levels and weight were detected every 2 weeks. The excision of rats’ renal was finished 4 weeks later. The pathological changes in renal tissues were examined by optical microscopy and electron microscope, and the expression levels of CXCL10/ CXCR3 were detected by RT-PCR. Results: Compared with the DM group, the renal injury, showed by electron microscope, was lessened significantly; the expression of CXCR3, showed by the results of RT-PCR, was reduced effectively in rats’ renal of the Puerarin group (p < 0.05), but the decrease of CXCL10 was not significant (p > 0.05). Conclusion: One possible reason, for lessening injury of DM rats’ renal by Puerarin, is that Puerarin relieves the inflammation by reducing the expression level of CXCR3.
